PENGARUH INDIKATOR BROMOTHYMOL BLUE DENGAN BROMOCRESOL PURPLE TERHADAP PIGMENTASI Trichophyton mentagrophytes PADA MEDIA SEREAL AGAR

Abstract

Trichophyton mentagrophytes menghasilkan metabolit alkali, sehingga ketika dikultur pada media yang mengandung indikator akan memberikan pigmentasi. Penelitian ini bertujuan untuk mengetahui perbandingan penambahan indikator bromothymol blue dengan bromocresol purple pada media sereal agar dengan melakukan studi pigmentasi Trichophyton mentagrophytes. Penelitian ini dilakukan dengan cara menambahkan indikator bromothymol blue dan bromocresol purple dengan variasi konsentrasi 2,2%, 1,6% dan 1% pada media sereal agar. Kemudian dilakukan isolasi Trichophyton mentagrophytes dengan metode single dot dan diinkubasi pada suhu 25-28°C serta dilakukan pengamatan pigmentasi selama 7 hari. Setelah melakukan penelitian, data dianalisis dengan uji Kruskal-Wallis, menunjukan adanya perbedaan signifikan pada penambahan bromothymol blue dengan variasi konsentrasi 2,2%, 1,6% dan 1%  serta tidak adanya perbedaan signifikan pada penambahan bromocresol purple. Dari hasil tersebut dapat disimpulkan bahwa bromocresol purple lebih cepat memberikan pigmentasi dibandingkan dengan bromothymol blue, hal ini dikarenakan bromocresol purple memiliki range pH yang lebih pendek yaitu 5,2 – 6,8 dibandingkan dengan bromothymol blue yaitu 6,0 – 7,6.
