EKSTRAKSI DAN PURIFIKASI FUKOSANTIN DARI RUMPUT LAUT COKELAT Sargassum sp. SEBAGAI ANTIOKSIDAN

Abstract

Fucoxanthin is one of the pigments that were produced in carotenoid biosynthesis which contributed more than 10% of the total carotenoid in nature, one of them was found in brown seaweed Sargassum sp. This is a type of brown seaweed that distribution is quite dominant at the Kalianda Lampung beach. The aim of this research was to purify fucoxanthin from brown seaweed Sargassum sp. which has potential as an antioxidant agent for the health sector. The research design is experimental with the research sample being Sargassum sp. brown seaweed. The extraction method used in this research was single maceration. The ethanol crude extract obtained was used for the phytochemical assay, then continued to purify process by using silica gel chromatography open column. Purified fucoxanthin content was analyzed with thin layer chromatography (TLC) and high-performance liquid chromatography (HPLC). Purified fucoxanthin was tested for antioxidant activity with 2,2-Diphenyl-1-picrylhydrazy (DPPH) method. The result of the phytochemical assay showed that ethanol crude extract Sargassum sp. contains active compounds such as alkaloid, flavonoid, phenolic, triterpenoid, and saponin. The result of the TLC analysis showed that the fucoxanthin fraction purification result obtain the main spot with Rf was 0,5, and the result of the HPLC analysis obtain fucoxanthin content was 0,59 mg/g. Antioxidant activity from fucoxanthin fraction had IC50 value = 87,64 ppm. This showed that Sargassum sp. active fraction has potential in the health sector as a natural antioxidant.
