PEMANFAATAN EKSTRAK BUNGA ASOKA SEBAGAI ALTERNATIF PENGGANTI EOSIN PADA PEWARNAAN HEMATOXYLIN EOSIN
Abstract
Preparation of tissue preparations using hematoxylin eosin staining. The price of reagents used is relatively expensive so it has the potential to use local raw materials to replace eosin staining. Red Ashoka flowers (Ixora coccinea) contain red anthocyanin glycosides that can be used as an alternative to eosin. The research aimed to utilize environmentally friendly Ashoka flower extract as a substitute for eosin in hematokxylin eosin staining. The research design used is true experiment design. Sampling techniques in research using nonprobability in the form of purposive sampling. Samples of human breast tissue (mammae) be rjumlah 32 tissue preparations to see the results of the examination using Ashoka flower alternatives as many as 16 preparations and eosin as a control as many as 16 preparations. The examination data will be processed by descriptive analysis based on the results of the assessment score of tissue preparation. In hematoxylin eosin staining control 100% showed very good preparation quality with a score of (4), producing blue in the cell nucleus is very clear, pink is very clear, and can be diagnosed, while for the ash flower extract staining 93.8% showed less good preparation quality with scores of (2) and (1), producing blue in the cell nucleus, less pink but still can be diagnosed. Chi-Square test results obtained p-value 0.000<0.05 shows that there are significant differences from the two staining processes using Ashoka flower extract with eosin.
