UJI KEPEKAAN ANTIBIOTIKA Staphylococcus aureus DAN Escherichia coli PADA MEDIA TAHU PENGGANTI MUELLER HINTON AGAR
Abstract
Mueller Hinton Agar is recommended medium by WHO for testing the sensitivity of bacteria to antibiotics while Tofu Media can generally be used as a basal as well as nutrient agar. This study aims to determine the differences in the results of the sensitivity test of Staphylococcus aureus and Escherichia coli using Mueller Hinton Agar and Tofu Media. The susceptibility test research used Kirby-Bauer diffusion method, where each bacterium was tested against 5 types of antibiotics. Staphylococcus aureus was tested against Trimethoprim-sulfamethoxazole, Tetracycline, Clindamycin, Vancomycin, and Erythromycin while Escherichia coli was tested against Ampicillin, Tobramycin, Amoxiclav, Ciprofloxacin, and Nalidixic Acid. Bacterial suspension with a concentration according to the standard Mac Farland 0.5 was planted on Mueller Hinton Agar and Tofu Media, then an antibiotic-disc was placed. The zone of inhibition was seen as a clear zone around the antibiotic-disc. The results showed that the inhibition of antibiotics TE 30 g, MEM 10 g, and OX 5 g against S. aureus in tofu media was greater than that of MHA, while SXT 25 g and E 15 g were less inhibited. The inhibitory power of AMP 10 g and AML 25 g antibiotics against E. coli in tofu media was greater than that for MHA, for C 30 g was the same, while TOB 10 g and CIP 5 g were smaller. The conclusion of the study showed that Tofu media could be used as a substitute for MHA in the sensitivity test of S. aureus and E. coli.
