PENGARUH LAMA SIMPAN DAN KONSENTRASI MANITOL PADA PACKED RED CELL SAGM TERHADAP KADAR HEMOGLOBIN DAN HEMATOKRIT

Abstract

Storage of Packed Red Cells can cause biophysical and biochemical changes, these changes include a decrease in the stability of erythrocytes as measured by the release of hemoglobin and can affect the hematocrit value. The addition of preservatives is intended to prevent blood clots and maintain cell function. With the addition of an additive solution, it can supply extra volume and nutrients for longer storage and improved flow with the addition of an additive solution. SAGM, which is made up of saline, adenine, glucose, and mannitol, is a popular additive solution. The lysis of erythrocytes can be minimized by adding 1-2% mannitol which can reduce lysis by up to 50%. Erythrocytes will be able to be kept for up to 42 days with the help of the additional solution. The purpose of this study is to see how storage duration in Packed Red Cell SAGM affected hemoglobin and hematocrit levels, as well as how mannitol concentration in Packed Red Cell SAGM affected hemoglobin and hematocrit levels. Treatments for storage period of Packed Red Cell (PRC) SAGM for 0 days, 14 days, and 28 days, and changes in mannitol concentrations of 0.525 percent, 1 percent, and 1.5 percent on hemoglobin levels and hematocrit values, were provided in a completely randomized design. The research data obtained were analyzed statistically using the General Linear Model Repeated Measures Two Way test, the results of the examination of hemoglobin levels and hematocrit were Asymp values. Sig > 0.05. As an end result, it can be stated that storage duration for Packed Red Cell SAGM and mannitol concentration have no effect on hemoglobin and hematocrit levels.
